Uncovering the lymphatic uptake, pharmacokinetics, and biodistribution of mRNA-lipid nanoparticles after intravenous and subcutaneous injection

Using three established ionizable lipids, researchers measured RNA, lipid components, lymphatic transport, plasma exposure, and tissue reporter expression in rats. After subcutaneous injection, lymph concentrations exceeded plasma concentrations, but lipid and RNA trafficking did not fully match. The study demonstrates why tracking a lipid label alone may not accurately represent intact RNA delivery or productive expression in tissues.

Programmable mRNA vaccine materials for temporal control of antitumour immunity

A Perspective proposes designing mRNA vaccine materials around changing immune conditions before and after cancer surgery. The framework considers a broadly applicable preoperative prime followed by a personalized postoperative boost, with carriers controlling expression and immune activation. It is a conceptual design roadmap, not evidence from a new clinical trial.

MAGEA11 recruitment enables selective BET degradation in cancer cell models

Researchers developed BET-targeting PROTACs that recruit the tissue-restricted scaffolding protein MAGEA11 rather than relying on widely expressed ligases. Lead compound 105B degraded BET proteins in two cancer cell lines but not in HEK293T cells. Mechanistic experiments implicated MAGEA11 and HUWE1. The authors emphasized that further optimization is needed to produce selective cytotoxicity.

MEPSGEN launches small-volume NanoCalibur Mini nanoparticle formulation system

MEPSGEN has launched NanoCalibur Mini for automated formulation of lipid and polymeric nanoparticles at volumes of 0.1 to 3 milliliters. The system targets early screening when payloads and excipients are scarce. The company says its platforms retain identical microfluidic mixing geometry across scales and increase throughput through parallelization, an approach intended to reduce process redevelopment during technology transfer.

WuXi AppTec reports nine-month 2026 results and raises full-year guidance

WuXi AppTec reported nine-month 2026 revenue of RMB 45.54 billion, up 38.6% year over year. Revenue from continuing operations rose 46.3%, and continuing-operations backlog reached RMB 76.74 billion, up 36.4%. The company also raised full-year guidance. Total-revenue and continuing-operations growth rates use different reporting scopes and should not be treated as interchangeable.

Moderna

INTerpath-001 details Phase 3 benefit of personalized mRNA therapy in melanoma

Moderna reported detailed results from the randomized, double-blind Phase 3 INTerpath-001 trial in completely resected stage IIB–IV melanoma. Adding intismeran autogene to pembrolizumab improved recurrence-free survival in the prespecified intention-to-treat analysis: hazard ratio 0.58 (95% CI 0.45–0.75), with 24.6 months median follow-up. Grade 3–5 treatment-related adverse events occurred in 14.2% versus 13.4%. The treatment remains investigational.

Vitamin B6 ternary eutectics change solubility and permeability in two BCS IV drugs

Researchers combined pyridoxine hydrochloride with mesalamine or acetazolamide and p-aminobenzoic acid to prepare ternary eutectic solids. The systems altered pH-dependent solubility and membrane permeability. Characterization distinguished a physical mixture from a cocrystal-containing eutectic, emphasizing that multicomponent formulations can have different phase identities. The reported laboratory results do not establish human bioavailability improvements.

Ivacaftor study links milling-induced solvates to starting form and crystal surfaces

A study of ivacaftor examined how solvent identity, liquid-additive volume, milling time, and starting solid form affect mechanochemical solvate formation. Crystalline Form C and amorphous material followed different transformation patterns. Structural and surface analyses suggest that mechanically exposed polar groups contribute to solvent incorporation, rather than solvent class or apparent dissolution capacity alone explaining the outcome.

A Scalable, Antibody-Free Lipid Nanoparticle Achieves Preferential mRNA Delivery to Human HSPCs In Vivo Without Conditioning

A ligand-free nanoparticle formulation preferentially delivered mRNA to human hematopoietic stem and progenitor cells in humanized mice without conditioning. It reduced hepatic delivery relative to an SM-102 comparator and enabled functional Cas9-mediated editing of human CD34-positive cells. Single-cell analyses supported tolerability under the conditions examined. Durable stem-cell function and clinical delivery still require further investigation.

Supramolecular gel supports furosemide salt-cocrystal growth at lower concentration

Researchers developed a calix[4]arene-based supramolecular gel whose assembly is triggered by dihydrogen phosphate. A salt cocrystal containing furosemide and its tetrabutylammonium salt grew within the gel at a lower furosemide concentration than required in the corresponding solution. The work demonstrates a crystallization environment, rather than a clinically validated pharmaceutical formulation or a ready-to-use manufacturing process.

Enlicitide analysis examines LDL lowering across kidney function groups

A Kidney Week report described pooled CORALreef trial findings for oral PCSK9 inhibitor enlicitide across kidney function groups. Participants with estimated glomerular filtration rates below 60 had a 65.9% placebo-adjusted LDL cholesterol reduction at Week 24. This subgroup analysis addresses lipid lowering; it does not establish fewer cardiovascular events, which require dedicated outcomes evidence.

Asymchem reports nine-month revenue growth of 16.52%

Asymchem reported consolidated revenue of RMB5.395 billion, up 16.52%, and attributable net income of RMB852.2 million for the nine months ended September 30, 2026. Attributable net income grew 6.49%, trailing the increase in revenue over the same nine-month period. The quarterly filing is unaudited and covers the year-to-date period.
